We constructed a comparative single-cell transcriptomic map of regenerating and non-regenerating frog tadpole limbs, showing that regeneration reuses some of the limb-development–related cell populations. We further demonstrated that secreted inhibitory signals, arising from cartilage and bone, progressively restrict regenerative competence, reframing regeneration loss as an actively regulated process driven by the cellular environment.
